Only if Kami tells him about the weakness which he wouldn't know otherwise. Then it's Goku + Kami vs Superman. That's a different battle. Also, Goku's weakness would be getting punched once. He would probably just explode. And if it was a battle of endurance and wits. Superman can just take a nap in space. I hear Goku instantly dies there. I'd consider that a weakness.
