I generally have to hit the pause button, then play again to get it to start. Though it could be possible you need a Twitch account.

Unfortunately I am not the one to offer any real suggestions with troubles with Twitch.TV's player.

Use VLC for playing it (other programs might work too). I got the direct link by using this: http://www.fakegaming.eu/TwitchTV/download.php
